An Experimental study on the Malignant Fibrous Histiocytoma Induced by DMBA(9,10-Dimethyl-1, 2-Benzanthracene)

Abstract
To investigate the morphological characteristics and histogenesis of the malignant fibrous histiocytoma(MFH) induced by DMBA(9, 10-Dimethyl-1, 2-Benzanthracene), 0.1§¢ of 2% DMBA liquid paraffin solution was injected into the knee joint cavities
of
the
Sprague Dawley rats(3 times, 4 weeks interval), thereafter, histologic and enzyme histochemical staines were performed.
@ES The obtained results were as follows:
@EN 1. Incidence rate of tumors was 94% in DMBA treated group, the first masses were palpated at 10-16 weeks after DMBA injection, and expired at 24 to 33 weeks.
2. The size of the first palpated masses were 2.0¡¿1.4cm on the average, and size of the last palpated masses at death were 5.7¡¿3.8¡¿3.6cm on the average.
3. Grossly, most of the tumors developed single, and were nodular, hard, and well encapsulated,. The cut surface disclosed whitish yellow fibrous tissue, and necrosis, hemorrhage, and cyst formation were noted.
4. Microscopically, the findings were compatible with typical form of MFH, storiform-pleomorphic type. Most of the tissues revealed storiform, fascicular, and cartwheel arrangement of the plump fibroblasts, and pleomorphic histiocytes with
single
or
several nucleated giant cells, bizarre cells, vacuolated cells, and myofibroblast like cells. Frequent mitosis were shown. And locally, myxoid change, vascular proliferation, and severe inflammatory cell infiltration with necrosis were observed.
5. PAS positive cells with varing degree of stainity, collagen and reticulin fibers stained with trichrome, van Gieson, and reticulin stains were demonstrated. Myofibroblastic cells were stained with red color for trichrome stain, but cross
striations
were not observed in PTAH stain.
6. Acid phosphatase activity was strong in almost malignant cells, but alkaline phosphatase and nonspcific esterase activities were not recognized.
Above findings are compatible with malignant fibrous histiocytoma and suggest that this tumor originated from undifferentiated mesenchymal cells having the capacity for differentiation toward histiocyte and fibroblast.
